This online real-time presentation focused on the applications of siRNA to identify and inhibit clinically relevant RNA targets. Emphasis will be placed on challenges involved in development of siRNAs as viable therapeutic agents in both systemic and local delivery settings. Most eukaryotic cells contain an intracellular defense system against invasion from outside agents and aberrant expression of endogenous genetic elements. This defense system is triggered by agent-specific double-stranded RNA, which is engaged by an RNAseIII-like nuclease called Dicer. Dicer processes dsRNA into short duplexes of 21-25 nucleotides in length. These short duplexes are recognized by a large protein complex called RISC, RNAi induced silencing complex. The RISC complex is activated in the presence of ATP and becomes capable of specific mRNA recognition guided by the sequence of one of strands of the short duplex RNA. The target mRNA is endonucleolytically cleaved leading to inhibition of expression at the translational level. Although the precise biochemical details remain to be worked out, the process of RNA interference (RNAi) mediated by short dsRNA (siRNA) has already proven to be of immense value in gene silencing in cell culture. Efforts are underway to extend the utility of siRNA into in vivo settings as a highly specific yet broadly applicable mechanism for gene silencing. These efforts require chemical modifications to stabilize siRNA against nucleolytic degradation and mechanisms to optimize pharmacokinetic performance and delivery to the target organ.
