Providing an overview of the newest data and technologies relating to the expression and characterization of proteins for biological understanding and therapeutics. One of the most fundamental approaches to understanding protein function is to correlate expression-level changes as a function of growth conditions, cell-cycle stage, disease state, external stimuli, level of expression of other proteins, or other variables. Although DNA microarray analysis offers a massively parallel approach to genome-wide mRNA expression analysis, there often is not a direct relationship between in vivo concentration of mRNA and encoded protein. Differential rates of translation of mRNAs into protein and of protein degradation in vivo are but two factors confounding the extrapolation of mRNA to protein expression profiles. Recent research, to be presented, shows that protein concentration can vary by >100-fold for a given mRNA concentration. In addition, microarray analysis cannot detect, identify, or quantify posttranslational protein modifications—key modulators of protein function. In measuring the level of the biological effector protein molecule, not just that of a protein’s message, protein expression analysis stands to advance significantly the contribution of proteomics to the development of novel therapeutics.
