This live webcast addressed the major challenges of setting design contraints in the tissue engineering of load-bearing orthopaedic structures to ensure long-term functionality of the constructs after surgery. Constructs must be both stiff and strong enough to tolerate the large and rapidly-applied loads experienced in vivo and yet not so stiff that cells are stress-shielded in the construct or joint kinematics are adversely affected by the presence of the implant. This virtual seminar will first focus on applying principles of functional tissue engineering (FTE) to better evaluate tissue engineered repairs using relevant in vivo forces and deformations as guides for cell and materials selection. Examples of repair strategies will be discussed including the use of stem cells to reduce or eliminate the need for harvesting autologous tissue that could compromise normal tissues and joints. The seminar will then discuss how applying in vivo mechanical signals in culture can precondition these constructs to their future environment in vivo. Finally, the seminar will examine how tissues of increasing structural complexity require increasing numbers of functional tissue engineering parameters (FTEPs) in order to improve the final design and the repair outcome after surgery.
